چکیده

The carbohydrate fraction of human milk includes a number of chemically defined oligosaccharides which contain the 6-deoxyhexose, L-fucose (2, 3). The present report describes a particulate enzyme from lactating canine mammary tissue’ which catalyzes the synthesis of fucosyllactose,2 the simplest member of this group, by transfer of L-fucose from guanosine diphosphate L-fucose to lactose. If fucosyllactose were formed by the preceding reaction in viva, the lactose moiety of the trisaccharide should arise from free lactose and, as a consequence, resemble it in 14C distribution after administration of a labeled precursor. This metabolic relationship was investigated in lactating dogs by modifying the technique used by Wood, Siu, and Schambye (5) in their extensive investigations of lactose synthesis by the bovine mammary gland. Glycerol-% was injected into the artery supplying the mammary gland, and the amount and distribution of radioactivity were determined in the oligosaccharides subsequently isolated from the milk. In addition to lactose and fucosyllactose, sialyllactose and a higher oligosaccharide fraction were isolated, and the specific activities of their component sugars were determined. The results indicate that n-glucose is the initial acceptor to which other sugars are sequentially transferred in the biosynthesis of fucosyllactose as well as other oligosaccharides found in milk.

Structural Basis for Norovirus Inhibition by Human Milk Oligosaccharides.

Histo-blood group antigens (HBGAs) are important binding factors for norovirus infections. We show that two human milk oligosaccharides, 2'-fucosyllactose (2'FL) and 3-fucosyllactose (3FL), could block norovirus from binding to surrogate HBGA samples. We found that 2'FL and 3FL bound at the equivalent HBGA pockets on the norovirus capsid using X-ray crystallography. Our data revealed that 2'FL ...

Determination of each neutral oligosaccharide in the milk of Japanese women during the course of lactation.

Using reverse-phase HPLC after pyridylamination, we quantified the concentrations of major neutral oligosaccharides in the milk of sixteen Japanese women collected at 4, 10, 30 and 100 d postpartum. In colostrum and mature milk (30 d lactation), lacto-N-fucopentaose (LNFP) I was the most abundant oligosaccharide, followed by 2'-fucosyllactose (2'-FL) + lacto-N-difucotetraose (LNDFT), LNFP II + ...

Human Milk Oligosaccharides Influence Neonatal Mucosal and Systemic Immunity.

The immune system of the infant is functionally immature and naïve. Human milk contains bioactive proteins, lipids, and carbohydrates that protect the newborn and stimulate innate and adaptive immune development. This review will focus on the role human milk oligosaccharides (HMO) play in neonatal gastrointestinal and systemic immune development and function. For the past decade, intense resear...

The principal fucosylated oligosaccharides of human milk exhibit prebiotic properties on cultured infant microbiota.

Breast-fed infant microbiota is typically rich in bifidobacteria. Herein, major human milk oligosaccharides (HMOS) are assessed for their ability to promote the growth of bifidobacteria and to acidify their environment, key features of prebiotics. During in vitro anaerobic fermentation of infant microbiota, supplementation by HMOS significantly decreased the pH even greater than supplementation...

Temporal Change of the Content of 10 Oligosaccharides in the Milk of Chinese Urban Mothers

Breastfed infants tend to be less prone to infections and may have improved cognitive benefits compared to formula-fed infants. Human milk oligosaccharides (HMO) are the third most abundant component of human milk, but are absent from formulae. They may be partially responsible for the benefits of breastfeeding. In this cross-sectional observational study, the HMO composition of milk from Chine...
